In this study, Marino et al. revealed that the sodium-myo-inositol cotransporter 1 (SMIT1), a member of the sodium/glucose co-transporters (SGLT) family, facilitates myo-inositol uptake into cardiac myocytes, promoting Na+ accumulation and inositol 1,4,5-trisphosphate (IP3)- and Ca2+-dependent pro-hypertrophic signalling. Deletion of SMIT1 protects from heart failure through upregulation of carabin, which inhibits calcineurin/NFAT and Ras/ERK1/2 signaling to prevent maladaptive remodeling. To understand why SMIT1 may be a novel therapeutic target to treat heart failure, read the full study. Cytosolic calcium handling signature: integration with clinical predictors enhances prediction of post-operative atrial fibrillation  Atrial fibrillation (AF) begets AF – this famous paper by Maurits Allessie* coined a concept in which electrical remodeling induced by AF predisposes to develop AF. A key factor of this remodeling is downregulated calcium currents. In this study, Fakuade et al. reveal that in atrial myocytes of patients with sinus rhythm who develop AF after cardiac surgery, calcium transients are already reduced when they enter cardiac surgery, and this defect predicts post-operative AF risk and enhances clinical risk prediction scores.
